What Is SPOT-MAS?

SPOT-MAS (Signal of Tumour by Methylation, Amplification and Sequencing) is a next-generation cancer screening blood test. It analyses circulating tumour DNA (ctDNA), which are tiny fragments of genetic material that tumour cells shed into the bloodstream. Using DNA sequencing and a machine-learning algorithm, SPOT-MAS detects cancer-associated signals across multiple organ systems simultaneously.

This is not a diagnostic test and it does not confirm cancer. A positive signal means there is an indication that warrants further investigation by our doctor. A negative result provides meaningful reassurance. Our doctors will explain exactly what your result means in the context of your personal health history.

What Does SPOT-MAS Screen For?

SPOT-MAS screens for early signals across 10 organ systems and over 75 cancer subtypes. These 10 cancers collectively account for more than 60% of cancer-related deaths in Southeast Asia.

  • Biliary Tract - Often asymptomatic until advanced stages
  • Breast - No.1 cancer in women in Singapore; early detection improves survival significantly
  • Cervical - Preventable and treatable with early detection
  • Colorectal - Among the most common cancers locally; highly survivable if caught early
  • Liver - Strongly linked to Hepatitis B, which remains prevalent in Singapore
  • Lung - One of the top causes of cancer death in Singapore; symptoms often appear late
  • Nasopharyngeal (NPC) - More prevalent in Southeast Asian populations than globally
  • Ovarian - Challenging to detect early; benefits greatly from proactive screening
  • Stomach (Gastric) - Higher incidence in Chinese Singaporeans; often diagnosed late
  • Thyroid - Increasingly common; excellent outcomes with early intervention

How Accurate is SPOT-MAS

SPOT-MAS is the first multi-cancer early detection test in Asia to complete a large-scale prospective clinical validation. The K-DETEK study, published in BMC Medicine, evaluated the test in over 9,000 asymptomatic adults and demonstrated the following:

  • 99.8% Specificity: Extremely low false positive rate - very unlikely to flag a signal when no cancer is present
  • 99.9% Negative Predictive Value: A negative result offers very high confidence that no cancer signal was detected
  • 78.1% Sensitivity: Detected cancer signals in approximately 4 out of 5 people who had cancer
  • 84.0% Tissue Origin Accuracy: When a cancer signal is detected, the test correctly identifies where it is coming from in 84% of cases

Who Should Consider SPOT-MAS?

SPOT-MAS is recommended for healthy adults aged 40 and above. It may also be appropriate for younger adults with known risk factors. Our doctors will assess your individual circumstances during the consultation before prescribing the test.

You may be a good candidate if you:

  • Are 40 years of age or older with no current cancer diagnosis
  • Have a family history of cancer - particularly if a first-degree relative has been diagnosed
  • Have a history of Hepatitis B or C infection (linked to liver cancer risk)
  • Are a current or past heavy smoker
  • Drink alcohol regularly and in significant amounts
  • Have tested positive for inherited cancer gene mutations such as BRCA1, BRCA2 or TP53
  • Want a more comprehensive approach to cancer surveillance beyond standard screenings such as mammograms or colonoscopies
  • Are proactive about your long-term health and want greater peace of mind

SPOT-MAS is not suitable if you:

  • Have been diagnosed with cancer within the past 5 years
  • Have highly suspected signs of cancer already under investigation
  • Have specific findings on imaging that require immediate diagnostic workup (e.g. lung nodules, liver lesions, polyps over 1cm)
  • Are currently pregnant
  • Have had a bone marrow transplant or whole blood transfusion in the last 3 months

How to Prepare for Your SPOT-MAS Cancer Screening

SPOT-MAS is a simple blood test. Our Doctor will draw a small sample of blood from your arm, the process takes only a few minutes.

No fasting required. You may eat and drink as normal beforehand.

Continue taking your regular medications as prescribed.

No other special preparation is needed.

Things to Take Note Before Your SPOT-MAS Test

SPOT-MAS screens for cancer signals in your blood. It does not directly diagnose cancer. If any signals are detected, further diagnostic investigations will be recommended.

This test does not assess inherited cancer risk from family members.

As with all laboratory tests, there is a small margin of error. Our doctors will counsel you on what this means for your results and the clinical implications before and after testing.

After Your SPOT-MAS Cancer Blood Test

Once your SPOT-MAS multi-cancer early detection blood test is completed, here is what you can expect:

How You Will Receive Your Results

  • Results are typically ready within 12 working days from the time your sample is received at the laboratory.
  • Results will be sent to you via WhatsApp or email. If a follow-up consultation is needed, our clinic will contact you directly to arrange this.

What Your Results Mean

  • No cancer signal detected: No cancer-related signals were found in your blood sample. While this is reassuring, it does not completely rule out cancer. Continuing with regular health screenings as recommended by your doctor remains important.
  • Cancer signal detected and localised: Our doctor will arrange a review session to go through your results in detail and refer you to the appropriate specialist to further investigate the area of localisation and confirm findings.
  • Cancer signal detected but not localised: Our doctor will arrange a review session to discuss your results. You will most likely be referred to an oncologist to determine the next steps, which may include further imaging such as a PET scan to localise the signal and perform more definitive testing.

Confidentiality

  • Your results are handled with the highest level of confidentiality at all times.

Next Steps and Ongoing Care

  • Abnormal findings may require additional investigations to confirm or rule out a diagnosis.
  • If specialist care is needed, our doctor will refer you promptly to a trusted oncologist or specialist.
  • Our team will guide you through every step, coordinating next steps, explaining results in plain language, and making sure you feel supported throughout.
  • Even with a normal result, routine health checks, age-appropriate screenings, and a healthy lifestyle remain an important part of your long-term wellbeing.

How SPOT-MAS Fits Into Your Health Screening

SPOT-MAS is designed to complement your existing health screenings, not replace it. Standard screenings such as mammograms, colonoscopies, Pap smears, and PSA tests remain important because they go deeper into their specific areas. What SPOT-MAS adds is a broad simultaneous sweep across multiple cancer types, including liver, gastric, and ovarian cancers, etc, where no established population-level screening programme currently exists.

Think of it as widening your net. A comprehensive approach to cancer surveillance might include SPOT-MAS alongside your routine health screening, particularly as you move through your 40s, 50s, and beyond, or if you carry known risk factors.

Our doctors are happy to review your existing screening history and help you build a personalised screening plan that makes sense for your age, background, and risk profile.

About SPOT-MAS Cancer Test

What is SPOT-MAS?

SPOT-MAS (Signal of Tumour by Methylation, Amplification and Sequencing) is a multi-cancer early detection blood test. It analyses circulating tumour DNA (ctDNA) fragments in your blood to screen for early signals of 10 cancers simultaneously, using advanced genomic sequencing and artificial intelligence.

How does SPOT-MAS work?

When cells, including tumour cells, break down, they release small fragments of DNA into the bloodstream. SPOT-MAS analyses these circulating tumour DNA fragments, looking for methylation patterns and fragmentomic signatures associated with cancer. A machine-learning algorithm then interprets the results to detect whether a cancer signal is present, and if so, where in the body it may be originating from.

What types of cancer can SPOT-MAS detect?

SPOT-MAS screens for signals across 10 organ systems and over 75 cancer subtypes: Biliary Tract, Breast, Cervical, Colorectal, Liver, Lung, Nasopharyngeal (NPC), Ovarian, Stomach (Gastric), and Thyroid. These cancers account for more than 60% of cancer-related deaths in Southeast Asia.

Who should consider getting SPOT-MAS?

SPOT-MAS is recommended for healthy adults aged 40 and above. It is particularly relevant if you have a family history of cancer, carry known risk factors such as Hepatitis B infection or a smoking history, or want a broader cancer surveillance option alongside your existing health screenings.

How accurate is SPOT-MAS?
SPOT-MAS has been validated in the K-DETEK study, a large prospective trial of over 9,000 asymptomatic adults published in BMC Medicine (2025). The test demonstrated 78.1% sensitivity, 99.8% specificity, and 84% tissue origin accuracy. Our doctors will explain what these figures mean in the context of your individual results.
Is SPOT-MAS covered by insurance?
SPOT-MAS is not currently claimable under Medisave. Coverage under private insurance varies by policy, we recommend checking directly with your insurer before booking.
Where can I get SPOT-MAS in Singapore?
SPOT-MAS is available at both Regis Medical clinics, Holland Village (255 Holland Avenue) and Katong (437 Joo Chiat Road). The test must be prescribed by a doctor, so a consultation is required.
Is there any special preparation required before SPOT-MAS?
No fasting is required. You may eat, drink, and take your regular medications as normal before the test.
What happens if SPOT-MAS detects a cancer signal?
If the test detects a cancer signal, our doctor will arrange a review consultation to go through your results in detail. Depending on whether the signal is localised or not, you will be referred to the appropriate specialist or oncologist for further diagnostic investigations. A positive signal does not confirm a cancer diagnosis, it indicates that further investigation is needed.
Can SPOT-MAS be used for screening in healthy individuals?
Yes. SPOT-MAS is specifically designed for asymptomatic, healthy adults. The purpose of early detection is to identify signals before symptoms appear, when treatment outcomes are typically much better.
Is SPOT-MAS suitable for monitoring cancer recurrence?
SPOT-MAS is not indicated for individuals who have been diagnosed with cancer within the past five years or who are currently under active cancer investigation. If you have a history of cancer, please speak with your doctor about the most appropriate monitoring approach for your situation.
Are there any limitations or risks associated with SPOT-MAS?
SPOT-MAS is a blood test with no significant physical risks beyond a routine blood draw. As a screening test, it has a small margin of error, it may not detect every cancer present (false negative), and in rare cases may detect a signal where no cancer is found (false positive). Our doctor will counsel you on these limitations and what they mean for your specific results.
Is the SPOT-MAS Blood Test suitable for detecting all stages of cancer?
SPOT-MAS is primarily designed for early detection in asymptomatic healthy adults. It is most effective when used proactively before symptoms appear. It is not indicated for individuals currently under active cancer treatment or diagnosis.
Can SPOT-MAS detect cancer at all stages?
SPOT-MAS is designed to detect cancer signals at early stages, when they are harder to identify through symptoms. In the K-DETEK study, sensitivity was 65.2% for early-stage cancers and 88.3% for Stage IIIA. Earlier detection generally offers better treatment options and outcomes, but the test does not guarantee detection at every stage.
Can SPOT-MAS detect cancer in its early stages?
Yes, early detection is the primary purpose of SPOT-MAS. The test is most valuable when used proactively in asymptomatic individuals, where identifying a signal early can make a significant difference to treatment outcomes.
Does SPOT-MAS have any age restrictions?
SPOT-MAS is recommended for adults aged 40 and above. Younger adults with significant risk factors may also be considered, our doctor will advise whether the test is appropriate for you.
Is SPOT-MAS approved in Singapore?
SPOT-MAS is processed at a MOH-licensed clinical laboratory. The test must be prescribed by a licensed doctor. Our doctors at Regis Medical will assess your suitability and prescribe the test following a consultation.
Does SPOT-MAS require repeat testing over time?
There is currently no universal guideline on testing frequency for multi-cancer early detection tests. Our doctor will advise on how SPOT-MAS fits into your long-term health screening plan and whether repeat testing at intervals makes sense for your risk profile.
Is SPOT-MAS suitable for all types of cancer?
No. SPOT-MAS screens for signals across 10 specific cancer types. It does not cover every form of cancer. A negative result does not guarantee the complete absence of cancer, and regular health screenings remain important.
What cancers does SPOT-MAS not detect?
SPOT-MAS does not screen for all cancers. Cancers not covered by the test include, but may not be limited to, prostate cancer, bladder cancer, pancreatic cancer, leukaemia, lymphoma, and skin cancers. Our doctor can discuss which additional screenings may be relevant for your individual risk profile.
What should I expect during the SPOT-MAS procedure?
Our doctor will first consult with you to assess your suitability and prescribe the test. You will then have a small blood sample drawn from your arm, a process that takes only a few minutes. Your sample is sent for analysis, and results are typically available within 12 working days.
Are there research studies supporting SPOT-MAS?
Yes. SPOT-MAS is backed by multiple peer-reviewed studies. These include a case-control study of 2,288 participants published in eLife, the K-DETEK prospective clinical trial of 9,024 participants published in BMC Medicine (2025), and real-world data from over 100,000 individuals across Southeast Asia. Real-world findings were presented at ESMO Asia 2025.
